PIXER®: A Breakthrough in Fluid Management

Pulsation at the pump outlet is a significant challenge for bioprocessing, disrupting electronic flow sensors and causing flow rate and pressure fluctuations. PIXER® eliminates these issues, ensuring smooth, precise fluid movement while optimizing efficiency. Designed for high-value biologics, it minimizes shear stress, operates at high pressures, and efficiently handles viscous fluids.

PIXER® ’s top inlet placement is a key mechanical innovation that allows direct positioning beneath a fluid reservoir, such as a bioreactor or retentate tank, in a tangential flow filtration (TFF) system. Unlike conventional pumps with side-positioned inlets, this vertical orientation leverages gravity for continuous fluid feed, enhancing suction efficiency and preventing air entrapment and cavitation—common issues in horizontal pump designs. This gravity-assisted design also reduces footprint and prevents tubing collapse under suction pressure, which is particularly beneficial for handling high-viscosity fluids or high flow rates.

PIXER® features a three, five, or seven-diaphragm radial pattern that minimizes hold-up volume, outperforming traditional linear diaphragm pumps. The design eliminates trapped air, facilitating quick purging while reducing shear stress on sensitive drug products such as monoclonal antibodies, proteins, and cells by ensuring uniform flooding of the diaphragms. This smooth, fluid entry results in a steady, low-pulsation outlet flow, preserving product integrity and enhancing performance with flow rates up to 200 liters per minute.

Another significant advantage is its multiple-outlet functionality. One outlet supports standard fluid transfer, while another recirculates fluid back to the vessel, maintaining homogeneity without requiring additional motors, shafts, or impellers. This streamlined approach reduces capital costs and eliminates the need for molded plastic covers for impeller rods, lowering footprint and quality control challenges. Many traditional systems require extra motors and impellers to ensure homogeneity, but PIXER® simplifies the process, reducing complexity and consumable costs.

Additionally, PIXER® delivers up to 50 percent more flow per RPM than competing diaphragm pumps, significantly improving energy efficiency and reducing power consumption in large-scale manufacturing. This efficiency translates to substantial energy savings, contributing to sustainability initiatives while maintaining peak performance. Rethinking Buffer Management: Inline Buffer Dilution

Continuing its tradition of customer-centric innovation, Alphinity tackled the challenges of traditional buffer preparation faced by a leading biopharmaceutical manufacturer. Moving large buffer totes consumed valuable floor space and introduced logistical complexities that slowed production. Alphinity’s buffer dilution skid offered an immediate solution, enabling ondemand, point-of-use buffer production.

Building on this success, the manufacturer embraced Alphinity’s inline buffer dilution system, which integrates seamlessly with existing Water for Injection (WFI) infrastructure. The system dramatically reduced water usage and storage requirements by using six single-use bags filled with highly concentrated buffer stock (at 10x concentration). Operators select the desired buffer, specify the total volume, and initiate the process via an intuitive interface. The system then automatically verifies material availability and precisely mixes the concentrate with WFI to produce a ready-to-use 1x solution.

This innovation streamlines buffer preparation and simplifies downstream processes such as chromatography. Concentrated buffers are fed directly into the chromatography skid for realtime dilution and seamless transitions between different buffers, optimizing overall production efficiency, reducing energy consumption, and cutting operational costs.
